Magnesium glycinate is a chelated form of magnesium in which magnesium is associated with glycine, an amino acid naturally found in the body.Magnesium itself is involved in hundreds of physiological processes, including nerve function, muscle function, energy metabolism, and neurotransmitter regulation. Its biological role has also led researchers to investigate its relationship with sleep and relaxation.One proposed mechanism involves the interaction between magnesium and pathways related to neuronal excitability. Research on magnesium and sleep has been growing, although the evidence is not yet as consistent as it is for some better-established sleep ingredients.A systematic review and meta-analysis of three randomized controlled trials in older adults found that oral magnesium supplementation was associated with a reduction in sleep onset latency of approximately 17 minutes compared with placebo. However, the researchers rated the overall quality of evidence as low to very low and noted limitations in the available studies.A 2024 systematic review examined 15 interventional studies investigating magnesium supplementation for anxiety and sleep-related outcomes. Five of the eight studies that assessed sleep outcomes reported improvements in at least one sleep parameter, while others showed no improvement or mixed results. The authors emphasized that differences in dosage, formulation, study duration, and participant characteristics make firm conclusions difficult.More directly relevant to magnesium glycinate, a 2025 randomized, double-blind, placebo-controlled trial enrolled 155 adults with self-reported poor sleep quality. Participants receiving magnesium bisglycinate providing 250 mg of elemental magnesium daily for four weeks experienced a greater reduction in Insomnia Severity Index scores than the placebo group. The effect was statistically significant but modest, and the researchers called for longer studies using objective sleep measurements.This recent study is important because it provides human clinical evidence specifically for magnesium bisglycinate, rather than relying only on research involving unspecified magnesium forms.At the same time, the broader evidence suggests that magnesium should not be positioned as a standalone solution for every sleep concern. Its potential appears more relevant to sleep onset, relaxation, and overall sleep support, particularly in people with lower magnesium intake or status.
For supplement formulators, choosing the magnesium form is an important part of product development.
Magnesium supplements differ in their chemical form, elemental magnesium content, solubility, absorption characteristics, and tolerability. A 2021 systematic review of magnesium bioavailability found that inorganic magnesium formulations generally showed lower bioavailability than organic forms, although absorption also depended on dose and individual factors.
