The skin is primarily composed of the epidermis, dermis, and subcutaneous tissue. As we age, these components gradually degenerate, with the dermis being the most affected. After menopause, women experience a sharp decline in estrogen levels, and within the first five years, the collagen in the skin reduces by 30%. Research has found that accumulated senescent fibroblasts in the skin lead to collagen degeneration, elastin destruction, inflammatory cell infiltration, fibrotic changes, and fat tissue atrophy through a series of actions. Clearing senescent cells might be a new therapy for skin anti-aging.
On September 22, a study by Professor Kento Takaya's team from the Department of Plastic Surgery at Keio University School of Medicine in Japan, published in the journal Biogerontology, found that fisetin, a plant pigment commonly found in strawberries, apples, persimmons, onions, and cucumbers, can clear senescent cells in the skin. Researchers first induced senescence in cells using specific methods, then treated non-senescent (proliferating cells) and three types of senescent cells with fisetin. The results showed that compared to the control group, fisetin treatment significantly reduced the number of β-galactosidase-positive cells, a marker of senescence, and decreased the expression of senescence-related proteins p16 and p53, but no similar differences were observed in the fisetin-treated non-senescent cell group. These results indicate that fisetin can selectively clear senescent cells.
Furthermore, the expression of SASP-related molecular genes is elevated in senescent skin, and fisetin treatment significantly reduced their levels. These results suggest that endogenous senescence might be associated with elevated SASP levels. After removing senescent cells from aged human skin, the secretion of SASP also weakens. Fisetin can significantly improve age-related changes in the skin by selectively clearing senescent cells, making fisetin extract a potential new option for improving skin aging.
